
Lowenstein Jensen Medium, when supplemented with egg and glycerol, can be used for that cultivation, isolation, and differentiation of Mycobacterium spp. The initial formulation of Lowenstein Jensen media was as stated by Lowenstein who incorporated congo red and malachite eco-friendly to hinder undesirable bacteria 1,2. The current formulation relies upon Jensen's modification which eliminates congo red and utilizes a moderate power of malachite eco-friendly rather to avoid development of nearly all contaminants surviving purification from the specimen. Malachite eco-friendly functions being an inhibitory agent toward microorganisms apart from mycobacteria 3 as well as can serve as an inhibitor as well as pH indicator. This formulation also encourages the first possible development of mycobacteria. Fresh eggs are added aseptically for this media. During heating, the egg albumin coagulates, thus supplying a good surface for inoculation. Nitrogen, essential fatty acids, and proteins are provided by egg and asparagine. Glycerol works as a carbon source and it is favorable towards the development of a persons type tubercle bacillus while being unfavorable towards the bovine type. Don't add glycerol towards the medium if bovine or any other glycerophobic strains should be cultured. Formation of blue zone signifies home loan business pH by gram-positive contaminants (e.g. Streptococci) and yellow zones of dye destruction by gram-negative bacilli.

Look for indications of contamination and degeneration. Users of packaged media might be needed to do qc testing with a minumum of one known organism to show growth or perhaps a positive reaction and a minimum of one organism to show inhibition or perhaps a negative reaction (where relevant).
